The difference is noticable here. Using S3TC, scenes can have higher resolution textures with little performance hit. Alternatively, a 16Meg card with S3TC would give similar performance to a 32Meg card without S3TC. However, the texture-uploading bottleneck, which S3TC alleviates, is becoming a less critical obstruction to performance as the memory supplied on cards increases. Processing of geometry, triangle setup on the card and fill rate are the other major bottlenecks in the rendering pipeline. Scott says fill rate is reaching a point where it is no longer an issue.
